Great question. "5' & 3'" means five prime and three prime. Have you ever seen DNA's molecular structure? Well, these numbers indicate the carbon on the sugars that DNA has.

-The 5' bears a phosphate group ([tex]PO_{4} ^{-3}[/tex]), and the 3' bears a hydroxyl group ([tex]OH^{-}[/tex]).
